Mouse monoclonal antibodies We need about 1mg of antigen for the procedure. Minimum of 0,1 mg and best up to 5 mg. Development of hybridoma cells producing specific antibodies. You can order each step separately.
Step1 Immunizations and Titre Monitoring Immunization of 5 balb/c mice with different antigen doses to have a broader spectrum of responses to the antigen. After the first Elisa test we start with the fusion or do more immunizations, depending on the measured titre. This step needs 4 to 8 weeks.
Step2 Isolation of spleen cells from immunized mice. Direct use of spleen cells or cryoconservation of spleen cells. Frozen spleen cells can be used later for fusion with the same yield of positive clones compared to fresh fused cells. We can also send the spleen cells in this stage to the custmer lab for a self made fusion or for RNA extraction. This step needs two days.
Step3 Fusion of hyperimmunized spleen cells from one mice with a myeloma cell line. We can also send the fused cells to the custmers lab for self made screenings and clone isolations. This step needs two days.
Step4 Screening for positive clones by elisa method. Isolation of positive clones. Storage of positive clones in liquid nitrogen until sending to the customer. This step needs 2 to 3 months.
Step5 Subcloning to make shure that the antibody producing clones are monoclonal and /or to clone for highest producing cell subclones. This step needs 1-3 months.
